Accumulation of losses of heterozygosity and multistep carcinogenesis in pulmonary adenocarcinoma.

چکیده
Sixty-six replacing growth-type early lung adenocarcinomas, measuring 2 cm or less across their greatest dimension, were used to investigate allelic losses at eight loci on the eight chromosomes carrying the principal cancer-associated genes. In total, 2 (16.7%) of 12 type A tumors (localized bronchioloalveolar carcinoma, LBAC) and 11 (39.3%) of 28 type B tumors (LBAC with alveolar collapse), which correspond to early lung adenocarcinomas including cancers in situ, showed allelic losses in one or more of the regions examined. In contrast, 25 (96.2%) of 26 type C tumors (LBAC with active fibroblastic proliferation), which correspond to small but advanced tumors, showed allelic losses in one or more regions. The change in histology from type A to type C was characterized by a significant rise in the incidence of allelic losses (P < 0.01). Deletions of 3p, 17p, 18q, and 22q increased significantly during malignant progression. In type C tumors that showed heterogeneous histological features, the tumor cells in the central fibrotic areas exhibited more allelic losses than those in the peripheral bronchioloalveolar growths and were, therefore, considered to have progressed to a more advanced stage than the tumor cells in the peripheral regions.

Frequent loss of heterozygosity on chromosomes 1q, 5q, and 17p in human gastric carcinomas.
Recently, loss or inactivation of genes at specific chromosomal loci has been considered to be one of the important mechanisms during the development of human tumors. In order to identify tumor suppressor genes for gastric carcinoma, we performed restriction fragment length polymorphism analysis on 48 human gastric carcinomas. Allele losses were investigated for 14 specific loci on chromosomes ...
متن کاملHistopathological study of naturally occurring ovine pulmonary adenocarcinoma in native goat in Khuzestan, Iran
Sheep (ovine) pulmonary adenocarcinoma (SPA), also known as ovine pulmonary adneomatosis (OPA),jaagsiekte and ovine pulmonary carcinoma (OPC) is a contagious lung tumor in sheep and, more rarely, ingoats. This study was carried out in Khuzestan, in the southwest of Iran for a period of six months. A totalnumber of 3985 native breed goats were clinically inspected and postmortem examination was ...
متن کاملClose Correlation between Telomerase Expression and Adenomatous Polyp Progression in Multistep Colorectal Carcinogenesis1
To investigate the role of telomerase in the multistep Colorectal carcinogenesis, we examined telomerase activity in 31 adenomatous polyps and 22 paired cancer-normal mucosa specimens from non-hereditary nonpolyposis colorectal cancer patients. Telomerase activity was detected in 18% of normal mucosa, 16% of small «1.0 cm) polyps, 20% of intermediate polyps, 71% of large (>2.0 cm) polyps, and ...

Ovine pulmonary adenocarcinoma (OPA) is a contagious and transmissible lung cancer of sheepresembling human bronchiolo-alveolar carcinoma. In the present study, lungs of 9400 sheep slaughtered inFars province, Iran were examined morphopathologically. The OPA was diagnosed in the lungs of 21(0.22%) out of 9400 sheep. Frequency of involvement of different lobes in the affected lungs was includeda...
متن کاملTumorogensis : The Dual Role of Telomerase
Carcinogenesis is a multistep process characterized by the gradual accumulation of genetic changes that ultimately lead to cancer. These genetic mutations can impart limitless replicative potential to the cancer cells making them immortal. Telomeres are repeat nucleotide sequence TTAGGG that are present at the end of chromosomes. Its functions are to protect the chromosomal ends and to ensur...
